I searched around but didn't locate the exact details I need, so any input would help. This is my first attempt at reconstituting on my own and I want the steps confirmed to avoid irreversible errors. Should the bacteriostatic water and the dry peptide both sit at room temperature before mixing starts? If the water has been chilled or the peptide frozen, how long do they need to sit out? For the small 3 ml vials I'm using, the person who handled this previously used the same 30-unit insulin syringes I already inject with; is that acceptable or does reconstitution call for a different needle? When adding the water, do I follow the same air-draw and pull technique I use for injections, then swirl the vial, or is the process different?

You're heading in the right direction. Let both items reach room temperature first: take the water out for roughly five to ten minutes and the peptide for twenty to thirty if it was frozen; this reduces clumping and supports even dissolving. The insulin syringes work fine on small vials as long as you push the liquid in slowly to limit foam. For thicker material some switch to a bigger needle only for adding the water. Draw the needed volume, then ease it down the inner wall of the peptide vial instead of straight onto the powder. Wipe every stopper with alcohol beforehand, keep the vial upright when pulling doses later, date the vial after mixing, and refrigerate anything not used right away.

I never refrigerate my bacteriostatic water at all; it stays in a drawer so it is already at room temperature whenever I need it.

Thanks, that helps. One basic question though: when you say keep the vial upright while drawing, does that mean the vial sits flat on the table with the needle above it so I pull upward, or am I supposed to hold the vial straight above the needle and draw downward? I've been doing the second method so far.

I pull upward with the vial standing on its own; I just hold it steady in one hand and keep the needle above the stopper. Both directions are possible, but the upward draw feels more natural to me.

That is what I plan to do from now on, but the first batch I received came already mixed along with extra water, all packed together in one plastic tray. Not knowing any better I placed the whole tray in the fridge, so right now I have one opened water vial and one unmixed peptide vial sitting there together.

It really comes down to preference; some keep the water in a drawer while others, including me, store it in the fridge.

For the brand the manufacturer recommends room temperature storage at all times. With other brands the guidance after first puncture is less clear, so people end up storing it either way.

Which needle length do you use once the liquid level drops and you have to reach the bottom? Any tips for pulling out the last small amount left in the vial?

I flip the vial, look inside, and stop the needle tip just above the rubber stopper. If that still doesn't work you can get a longer needle that actually reaches the bottom.

It is not important at all; just choose whatever position feels easiest. Tilting the vial a little is fine if it helps.

I find it simpler to turn the vial upside down and only barely pierce the stopper; that speeds up the draw. When almost nothing is left I first push in a full syringe of air so vacuum does not slow me down. Several sources suggest chilling both the peptide and the water for an hour before mixing, though I keep my water at room temperature until an hour beforehand and then move it to the fridge. After the first puncture I do store it cold simply because it is convenient to keep everything together. I doubt the temperature choice makes any real difference.

A quick note on 10 ml vials: the stoppers on those often leave pockets that trap liquid. In that case I remove the crimp and stopper with a bottle opener after mixing, pull the liquid with a longer 27-gauge needle on a 3 ml syringe, attach a filter, and transfer the filtered liquid into a fresh sterile vial or cartridge.

The maker states not to refrigerate and to store at room temperature in a dark place because the alcohol becomes less effective when chilled.

I am simply following the printed directions. I have never run into any problems doing it this way.
