Has anyone used Semax or similar peptides for depression after SSRIs and herbal options didn't help? My wife has dealt with mild to moderate depression since middle school. SSRIs haven't worked well, so we're exploring other routes. If you've tried both antidepressants and peptides, how did they compare? Any suggestions on what to try or skip? Everyone's different, but real experiences would be useful.

No success with Semax for me, though I saw some mild improvement with PE 22-28.

I thought Semax was mainly for focus and mental clarity rather than mood. Selank seems to be the one aimed at mood and lowering anxiety. Anyone want to correct that?

Selank helped me, but I used it alongside an SSRI instead of replacing it. Ultimately switching SSRIs and adding an antipsychotic worked best. You could try it without much risk, though don't count on big results. Early effects can be placebo, so check if they hold up. Good luck.

I noticed a better mood and reduced depression. Started with nasal then switched to sub q. Some expect results in four days, but it took two weeks for me. Have you looked at P21?

There are tons of antidepressants besides just SSRIs.

True, there are many others. SNRIs and TCAs are off the table due to heart problems from an earlier eating disorder, and atypicals like Wellbutrin are also avoided for the same reason. So SSRIs have been the only option. The depression stays fairly mild and manageable, but we're still looking for extra support.

Other approved options still exist. Assuming you're in the US, several FDA-approved medicines fall outside the ones mentioned. Brains and science aren't limited to one country, and importing things unavailable locally is often straightforward and legal. Medications can do more than their approved uses. Contraindications aren't absolute. Earlier in my training we only had TCAs and MAOIs, and even post-MI patients could be managed with monitoring.

The contraindication concern seems overstated without knowing the exact heart issue. I'm on tricyclics despite heart disease. Avoiding bupropion over past history alone feels excessive if weight is stable now. There are SNRIs beyond SSRIs too. Some supplements show evidence for depression, including fish oil, l-methylfolate with SSRIs, vitamin D, l-carnitine, and PEA.

It feels like the peptide thread got turned into suggestions for prescription antidepressants. We've already researched this extensively with good doctors and a psychiatrist we trust. I came here specifically for peptide experiences after ruling out other meds. If you've used Semax, Selank or similar peptides for depression when standard treatments fell short, I'd like to hear about it.

Grateful for all the ideas and stories shared by others. Following additional investigation we've chosen to combine Selank with Semax and maybe P21 as well. I'm going to record our progress here hoping it might assist someone later on. We'll add them sequentially to spot any issues or adverse effects early on. Beginning with Selank at 250 micrograms injected under the skin we'll raise it to 500 if no problems arise. The same approach applies to Semax. Once we've finished two rounds of those we'll assess her emotional state and anxiety then decide whether to tweak things or start P21 through the nose. Sharing progress reports as we go for those who want to track along. With luck what we go through will benefit others thinking about this path.

My wife has had long-term anxiety and depression. Semax and Selank didn't do much, but oxytocin helped her feel more connected to herself and her priorities. We might try the first two again.

Not depressed right now, but have you considered a GLP-1? Reta gives me a steady sense of satisfaction and reduces cravings for other things.

Two-week update: Selank in the first week brought better sleep and calm without changing depression or anhedonia. Adding Semax in week two kept the calm and gave slight mood lift plus some GI upset that may be unrelated. We'll stay on the combo two more weeks then cycle off and try higher doses next round. Surprised so far but still watching for placebo effects.

With her low weight and eating-disorder background I'd be cautious about GLP-1s. I agree they can improve overall wellness though, as tirzepatide has for me.
