Summary

Personal experiences using Semax, Selank or similar peptides for depression when SSRIs and other standard treatments have failed

Users report mixed outcomes with Semax and Selank: some note mild mood or calm benefits while others see little change. Selank is viewed more for anxiety reduction and Semax for focus. The original poster rules out many other antidepressants due to heart and eating-disorder history and plans to trial a Selank-Semax-P21 stack while tracking effects. Several participants mention supplements or other peptides as possible additions.

Has anyone used Semax or similar peptides for depression after SSRIs and herbal options didn't help? My wife has dealt with mild to moderate depression since middle school. SSRIs haven't worked well, so we're exploring other routes. If you've tried both antidepressants and peptides, how did they compare? Any suggestions on what to try or skip? Everyone's different, but real experiences would be useful.

ClearKettle64archiveJul 23

No success with Semax for me, though I saw some mild improvement with PE 22-28.

PatientMeadow74archiveJul 23

What changes did you notice from the PE 22-28?

PatientTimber12archiveJul 23

I thought Semax was mainly for focus and mental clarity rather than mood. Selank seems to be the one aimed at mood and lowering anxiety. Anyone want to correct that?

KeenPebble33archiveJul 23

Selank helped me, but I used it alongside an SSRI instead of replacing it. Ultimately switching SSRIs and adding an antipsychotic worked best. You could try it without much risk, though don't count on big results. Early effects can be placebo, so check if they hold up. Good luck.

I noticed a better mood and reduced depression. Started with nasal then switched to sub q. Some expect results in four days, but it took two weeks for me. Have you looked at P21?

GreenAnchor65archiveJul 25

There are tons of antidepressants besides just SSRIs.

PatientMeadow74archiveJul 25
↳ replying to @GreenAnchor65

True, there are many others. SNRIs and TCAs are off the table due to heart problems from an earlier eating disorder, and atypicals like Wellbutrin are also avoided for the same reason. So SSRIs have been the only option. The depression stays fairly mild and manageable, but we're still looking for extra support.

GreenAnchor65archiveJul 25
↳ replying to @PatientMeadow74

Other approved options still exist. Assuming you're in the US, several FDA-approved medicines fall outside the ones mentioned. Brains and science aren't limited to one country, and importing things unavailable locally is often straightforward and legal. Medications can do more than their approved uses. Contraindications aren't absolute. Earlier in my training we only had TCAs and MAOIs, and even post-MI patients could be managed with monitoring.

CopperSignal27archiveJul 25

The contraindication concern seems overstated without knowing the exact heart issue. I'm on tricyclics despite heart disease. Avoiding bupropion over past history alone feels excessive if weight is stable now. There are SNRIs beyond SSRIs too. Some supplements show evidence for depression, including fish oil, l-methylfolate with SSRIs, vitamin D, l-carnitine, and PEA.

It feels like the peptide thread got turned into suggestions for prescription antidepressants. We've already researched this extensively with good doctors and a psychiatrist we trust. I came here specifically for peptide experiences after ruling out other meds. If you've used Semax, Selank or similar peptides for depression when standard treatments fell short, I'd like to hear about it.

PatientMeadow74archiveJul 25
↳ replying to @ClearKettle64

I'll check that out, thanks. We haven't tried P21 yet.

PatientMeadow74archiveJul 25

She already takes vitamin D and l-carnitine. PEA is new to me as a depression option. Thanks.

PatientMeadow74archiveAug 4

Grateful for all the ideas and stories shared by others. Following additional investigation we've chosen to combine Selank with Semax and maybe P21 as well. I'm going to record our progress here hoping it might assist someone later on. We'll add them sequentially to spot any issues or adverse effects early on. Beginning with Selank at 250 micrograms injected under the skin we'll raise it to 500 if no problems arise. The same approach applies to Semax. Once we've finished two rounds of those we'll assess her emotional state and anxiety then decide whether to tweak things or start P21 through the nose. Sharing progress reports as we go for those who want to track along. With luck what we go through will benefit others thinking about this path.

SteadyMarble20archiveAug 8

I'd like to see your updates.

CopperAlder51archiveAug 9

I'm about to start another P21 cycle. It works well with Semax and its versions. I found it helpful and would like to hear how it goes for you.

ClearLantern70archiveAug 9

My wife has had long-term anxiety and depression. Semax and Selank didn't do much, but oxytocin helped her feel more connected to herself and her priorities. We might try the first two again.

I ordered some of that last night after seeing it here.

CopperAlder51archiveAug 10

Mine is ready to ship once the test report comes through.

ClearKettle55archiveAug 12

Not depressed right now, but have you considered a GLP-1? Reta gives me a steady sense of satisfaction and reduces cravings for other things.

PatientMeadow74archiveAug 17
↳ replying to @PatientMeadow74

Two-week update: Selank in the first week brought better sleep and calm without changing depression or anhedonia. Adding Semax in week two kept the calm and gave slight mood lift plus some GI upset that may be unrelated. We'll stay on the combo two more weeks then cycle off and try higher doses next round. Surprised so far but still watching for placebo effects.

PatientMeadow74archiveAug 17
↳ replying to @ClearKettle55

With her low weight and eating-disorder background I'd be cautious about GLP-1s. I agree they can improve overall wellness though, as tirzepatide has for me.

IronCompass33archiveAug 17
↳ replying to @ClearLantern70

Really? Can you say more about the oxytocin? I'm in a similar situation and have been looking into it.

PatientBeacon57archiveAug 17

Unapproved and unstudied nootropics worry me because delivering compounds past the blood-brain barrier isn't simple, and the risks in this area often get overlooked.

ClearLantern70archiveAug 17

Describing this is difficult for me because I get nothing from it myself. My partner uses it in a tiny amount and right away feels affection along with a sense of calm and ease. She loses the urge to handle matters she can't influence. That's the way she talks about this item. Finding some isn't simple but the cost isn't high if it turns up.

RustQuill56archiveAug 17

I've been wondering if mushrooms came up as something she tried, since evidence points to benefits there. As for these compounds, anxiety has plagued me ever since my delivery five years back due to the trauma around childbirth. One gave slight relief. The other leaves me too wired so I've stayed away from it. Lately I've started checking out another that might suit better. Both can go under the skin via injection or get turned into nasal sprays by some, though any difference in results isn't clear to me and I've used the shots. Stronger help was what I expected from that one. Those classes of antidepressants did nothing either. At this point it feels like I'm defective with no way out of the endless pain.

SteadySparrow75archiveAug 18

Haha, that earlier remark of yours looks like it holds some groundless comedy, agree? 😎

CopperAlder51archiveAug 18

Semax gives me an energy boost but Adamax takes the intensity much higher. PE-22-28 came up in talks where some users mentioned improved mood and fewer depressive feelings even though human trials are missing. Oxytocin might fit in too. Since Selank works well for me I also tried the amidated NA version which feels more potent and keeps going longer.

PatientMeadow74archiveAug 18
↳ replying to @RustQuill56

Hadn't considered it, but her on shrooms might be fun. 😂

RustQuill56archiveAug 18
↳ replying to @PatientMeadow74

I meant to clarify sooner that this wasn't about consuming full mushroom doses but rather using very small amounts instead haha since these products appear through web shops in capsule versions or different formats. The one I had was a cocoa blend that went into my daily beverage and it made a noticeable improvement for some time.

PatientMeadow74archiveAug 18
↳ replying to @RustQuill56

Only joking around. 😉 Obviously this isn't about intense trips on psychedelic fungi. Might you mean lion's mane along with reishi? I came across certain beverage blends and soft chews containing those, where a couple folks shared encouraging feedback.

BlueThistle84archiveAug 18

I've never come across that pep from the sources I typically rely on, which makes me wonder if it's stocked anywhere else.

KeenBeacon78archiveAug 18
↳ replying to @BlueThistle84

My helpful bot explained that this compound matches the acetylated amidated version of selank. Multiple sellers in this community keep it available. As for the variant resembling that other product with an added adamantane group, I have not come across any offers of it.

CopperAlder51archiveAug 18
↳ replying to @KeenBeacon78

The genuine form of this substance corresponds to the matching item for the other one. It consists of the altered peptide featuring the attached cage at the terminus or the combined elements. Its weight ought to reach approximately nine hundred thirty units. Only a handful of individual sellers with vial and mist options have offered what seems authentic, leading me to skip buying any.

KeenBeacon78archiveAug 18
↳ replying to @CopperAlder51

It seems production might not happen if buyer interest stays too low. I really wish I could track down the genuine version.

RustQuill56archiveAug 18
↳ replying to @PatientMeadow74

Yeah you had a point actually haha it's the hallucinogenic type psilocybin. Roughly twelve months back I encountered folks taking small doses and the positive effects on their state of mind and feelings. I selected a company offering multiple formulations that also contain other types of mushrooms.

BlueThistle84archiveAug 19

During times of seasonal depression this makes quite an impact on how I feel. Applying it through the nose works well for me when I also increase my vitamin D.

CopperAlder51archiveAug 19

Hearing this from you marks the fifth RS to mention its helpfulness. A package is en route from the GB. Eventually.

I like the home grown version. The science is fun.

Using that compound ended up heightening my spouse's inability to feel any pleasure and people see varying effects from it.

For more than sixteen years those two prescriptions at the stated levels have kept my mood issues under control after the initial bipolar diagnosis shifted into major depression. This year hit with sudden deaths of four close relatives before summer closed out including my partner's dad during a standard heart procedure an aunt from hidden cardiac and pulmonary conditions a cousin whose cancer returned and my mother after abrupt intestinal blockage followed by infection stomach issues blood poisoning and finally breathing failure. She had shared my home for a decade. The losses dropped me into a deep clinical low I could not escape. Doses were already high and I raised one slightly yet the actual turnaround came from rotating two particular compounds alongside three workouts weekly plus a nasal spray each day. It lifted the mental haze let me sort through feelings and restored drive to leave bed and regain direction. My age in the late forties could matter here. I feel better now and intend to try one more item before sleep once it shows up.

BlueAlder13archive1d

My eighty-three-year-old mom receives one milligram of Selank every day for a month, then takes a month off. In the off month she gets one milligram of Semax along with one milligram of Pinealon. Selank maintains her cheerful disposition, whereas the other pair provides limited help against low mood yet mainly supports thinking abilities. Before leaving for my job each morning I come over to hand her the daily doses. Nothing bad can come from giving it a try. Wishing you success.

My relative tried those two compounds and said they seemed to ease her low mood. I have not used either one yet but expect to order them during my next gray market buy.
