I watched some peptide filtering videos and one on STG showed adding diluent to the filter first to wet it, letting it sit while getting everything else ready, then using it on the combined peptides. Has anyone else seen this step, and is it useful or worth doing? Filters have clogged or broken on me before, so I wondered if wetting them ahead of time might help.

Can't you just wet it using bac water? Draw 3 ml bac into a syringe, put 1 ml into the peptide vial, send 1 ml through the filter into a donor vial, pull the reconstituted peptide into another syringe and push it through the filter, then use the last ml of bac to rinse the filter.

Good question, I just got some filters myself. Pushing the liquid through was tougher than I expected.

Yeah, my venting filter needle stopped working on one vial and I lost 70 mg of glow.

Take it slow on the plunger. I use a longer needle on a 3 ml luer-lock syringe for drawing up or pulling from the vial so it reaches the bottom. After the solution is in the syringe with the filter attached, I switch to a shorter needle on the filter to push into the final vial. The longer needle also serves as the vent. Make sure the vent needle only just enters the vial to avoid backflow or foaming.

I tried the shorter needles for venting several vials and they must have dulled by the time I reached the pen cartridge. Now I just filter straight into the final cartridge the way the videos show, instead of filtering the bac and the source vial.

A piece of the stopper might have blocked the vent. Reusing it multiple times increases that chance each time.

Air couldn't escape through the vent needle. When I pushed the glow in through the other filter needle the rubber plunger popped out and everything leaked.

The only reason I can see for wetting a filter is to fill the empty space inside it with liquid. When dry it can hold onto 5-15 units like a sponge and throw off your dosing. Try practicing with bac water on an old vial first. I have never had a filter break or clog in a year of use. I stick with PES 0.22 µm 13 mm.

The first few times everything went fine. I use the smaller 4 mm ones and they cost more where I live so I try not to waste them. The issue was pointed out by others here: I didn't wait long enough for the peptides to settle and dissolve completely. Ten minutes turned out to be too short. Drawing and filtering some bac into the vial first sounds useful both to occupy the void space and to wet the filter if that helps. I need to reconstitute some soon so I want to get the process right this time.

I wet coffee filters for pour-over, backwash the pool sand filter when changing media, and flush the sediment and charcoal filters on my water system. So I plan to wet the peptide filter too. I want it to act as a one-way filter, so my idea is to draw bac water into the syringe, attach the filter, reconstitute the peptide, remove the filter to pull the solution out, put the filter back on, and then push into the final sterile vial.

You still need to flush afterward by pushing plain bac through as the last step, so split the bac into at least two portions. I got some 10 ml syringes for this so I can reconstitute three different vials at once with only one syringe and reduce how many times I poke the larger bac vial.

I think the idea of wetting first is that you won't need a flush afterward because the filter has only absorbed bac water, not any reconstituted peptide, the same way you wet a coffee filter. That said, doing the extra flush couldn't hurt.

But the filter still holds some liquid. If the peptide solution displaces the bac that was already in there, that amount gets lost. That's why the two-syringe three-injection method should work better and lets you do everything in the final vial without moving the filter on and off. Inject into the final vial through the filter, wet the filter with bac, push the peptide through, then inject more bac. You only swap syringes around.

In that case there is no real point to wetting, in my opinion. From what I have read, wetting standard PES syringe filters does not improve how well they work. The only real reason seems to be avoiding absorption, so if you are going to flush with bac at the end anyway then wetting first looks unnecessary.
