Eloralintide and Tirzepatide Rat Study

22 replies12 peopleJun 7, 2026☆ Follow
Summary

What does the rat study show about combining eloralintide with tirzepatide and what might stacking these pathways mean?

The thread covers ADA conference rat data where eloralintide added to tirzepatide produced additive weight loss, with adjunctive dosing after two weeks of tirzepatide. Participants note parallels to cagrilintide, discuss possible extension to retatrutide, and highlight longer appetite suppression as a key driver. They stress the data are preclinical only and raise questions around tolerability and lean mass loss at very high total reductions.

I came across this on a messaging channel. During a recent diabetes meeting a firm shared findings from an experiment with rodents that combined two compounds. The key point appears to be that their joint impact added up completely. That outcome fits what happens with another similar pairing. One notable detail in how the test was arranged involves starting with the first compound alone then layering on the second at different levels. The animals received the opening drug for a couple of weeks before the extra one was included. This approach matches what the firm likely expects in actual use where progress stalls on the initial medication and the follow-up one gets added later. The encouraging result is that further reduction in body weight showed up during that portion of the work.

IronLantern24archiveJun 7

I queried an artificial intelligence about outcomes from pairing these two compounds. Right now no human trials or animal studies cover their joint application. A logical guess can still be built from how each one functions separately, patterns seen with related pairings, and the overall development plan.

Projected results point to extra body weight reduction beyond either product alone. One already drives large drops while the second by itself reaches solid levels too. Combining something that heightens fullness and slows stomach emptying through a different route with a triple pathway activator ought to create complementary actions, echoing what animal work showed with a close match. This might allow bigger overall drops, break through stalls, or improve the ratio of fat versus muscle lost.

Fullness signals could strengthen further since the added route affects separate hunger pathways. Blood sugar handling, calorie use, and heart markers might improve as well, with the fullness component possibly helping keep muscle during loss.

Digestive side effects could build on each other, even though the newer option was created to cut nausea versus older versions. Slow dose ramps appear to hold problems in check based on prior paired tests.

The maker views the fullness agent as something to layer onto incretin medicines. Early safety checks with one such pair finished, mid stage tests continue with it, and later work examines adding it to steady incretin use for people who still carry extra weight.

Everything stays experimental without approval for combined use. Real results in people might differ and could bring stronger stomach trouble or other unknowns. Proper dose studies would be essential.

Overall the mix should produce stronger appetite control and weight reduction through separate routes, matching plans for greater effect when one agent alone proves insufficient.

I'd end up disliking meals forever after that haha

AmberSparrow63archiveJun 7

Early findings seem noteworthy. Pairing those two substances produced greater body weight reduction in overweight rodents than either one achieved separately. Extended hunger control played a major role in the outcome. The main drawback is that everything so far comes from animal tests instead of people. It still hints at the direction weight loss treatments are moving by hitting several mechanisms rather than depending on just one medication.

I see the point that the research so far involves just rodents, yet because one similar medication shows extra benefits when used by people, it's likely this new version will do the same for humans too. At least I hope so with a laugh. Picture what that could mean if the weight loss figures from each one were put together. That sort of result might really make a big difference against the widespread issue of excess weight.

Sure, supposing the findings from actual patients come anywhere near those figures, the impact might be enormous. That earlier compound has already transformed the field entirely. Layering in a powerful amylin mimic could drive reductions in body mass beyond anything previously achieved via pharmaceuticals. The critical factor remains how well folks handle it. Dropping such a large proportion of weight looks fantastic at first glance, but only if it doesn't bring nonstop discomfort, zero hunger, or excessive muscle decline. Yet succeeding in matching strong outcomes to fewer issues would offer a preview of the path ahead for obesity drugs. Crossing fingers the animal model pulls through.

That doesn't apply here at all. From the day I began this treatment I've seen myself as a rodent. 🐀 Right now I'm simply anticipating the next compound to finish my transformation.

CopperHarbour29archiveJun 7

She's definitely going to lose it once the launch takes place.

Outcomes beat anything from stomach operations. Such a pairing might finish off operative procedures for most situations. Absolutely sending good wishes toward the one leading this effort. He's showing the path forward for everyone else.

The link you shared caught my attention right away. Those two compounds caught my eye, yet the experiments with rodents really highlighted how they work together. I saved a picture of the graphs that demonstrate their combined impact in the animal tests.

I'm really keen on elora as well. The notion of alternative routes to tackle this issue and how this treatment might combine with current options is great. Appreciate sharing that image! What stands out most is the upper right section, especially the crimson line.

Looking at those charts more closely reveals some patterns worth noting. The combined dosing intake chart shows that elora tends to produce steadier daily food consumption compared to tirz or the pairing of both. Tirz leads to a pattern of one big eating day followed by two lighter ones, while elora shows far smaller swings from one day to the next. That three-day rhythm with tirz fits the every-third-day dosing schedule used here, and the steadier results with elora line up with its extended half-life keeping blood levels more consistent. On tirz by itself there were two days of strong suppression followed by a rebound where intake exceeded the untreated group. Once elora levels built up, those animals showed even tighter day-to-day consistency than the untreated ones. In the combined dosing chart the elora group drifted back toward the untreated intake level around the three-week mark, though the overall average stayed a bit below. That suggests anyone focused heavily on tracking calories might find elora easier to handle mentally than a GLP, unless they dose the GLP more often to reduce swings or make other adjustments. The study used the same chow throughout so it does not speak to those other adjustments. On the add-on chart the combination of tirz with the highest elora dose produced a sharp drop in appetite that lasted through two full dosing cycles. This happened even though tirz had already leveled off before elora was introduced, and the suppression came close to what was seen in the first two cycles of the combined-start chart. It does not appear to change the longer-term weight plateau, yet total intake ended up tracking in a very similar way for both the fresh GLP animals and the ones already used to it.

This stuff gets pretty close to what surgery can do all on its own. Once the four-way versions hit the market, I bet they'll do even more. Sure the downsides are noticeable, yet they beat out getting your belly cut open for good. My guess is after the three and four part versions gain traction, we'll move on to ones meant just for keeping things steady.

WarmMeadow92archiveJun 8

I have doubts on that notion. A more significant concern should be handled initially since this matches what the developers seem focused on. The key overlooked matter is how people aiming for larger body mass drops show lower success rates. The same pattern shows up in every similar treatment. Sorting out that aspect needs to happen ahead of anything else. One image displays the first compound while the next shows the alternative. Keeping results steady takes little effort beyond continuing with the drug.

Appreciate the research paper and chart being shared. For people thinking about trying the compound soon while prices stay high the upper right part of the chart gives some clues. The agent at the lowest amount tested showed almost no change yet ten times that produced five percent more body mass reduction and a hundred times the starting amount added ten percent further loss. Though this comes from animal work rather than human data it points to effects that do not increase in direct proportion to the amount used. If the biggest amount aims for ten percent loss by itself and lines up with the level of the other medication in the trial then even one tenth of it still gave five percent extra reduction suggesting smaller combined amounts could exceed what was anticipated assuming similar patterns occur in people. In addition if bigger amounts could tack on another ten percent beyond the other drug's results that would mark a major advance since total losses near forty percent could correct nearly every case except the worst excess weight and even milder results would still improve health substantially even with some weight remaining. I feel sympathy for the animals placed on the highest amount together with the other drug since they ate nothing for nearly seven days which lasts a long time for them. That situation surely involved negative reactions even if these creatures cannot regurgitate making symptoms tougher to spot yet it hints at problems from suddenly using a large amount of this agent while already taking the other medication possibly causing long stretches without food along with related issues that might need medical help such as fluids through a vein. This leaves me thinking careful handling of amounts matters for anyone testing the substance soon with gradual steps from small beginnings probably required.

Yeah, starting out at four hundred fifty when the goal is one hundred eighty means dropping one hundred fifty only covers part of the distance and leaves a hundred extra still hanging on. Aiming for two hundred seventy total through drugs looks incredibly tough, yet people figured the same thing about one hundred a couple years back, so who can say.

Dealing with someone at that high weight level looks difficult from what I know. Back in two thousand three my weight reached four hundred fifty five pounds. That same year I underwent stomach surgery which brought me down to two hundred forty five pounds. The result was remarkable and likely preserved my existence while achieving a forty six percent reduction in extra pounds. However this fell short of two hundred five pounds needed for standard ranges according to common standards. Beginning the medication at that point would have enabled me to attain the objective. Using it along with other similar compounds probably would have achieved an identical reduction to two hundred forty five pounds matching the operation outcome. This remains considerably below the standard range given my stature and build. This morning the scale showed two hundred fifteen pounds and the standard range appears reachable soon.

Way to go! Trying to find answers for the guy who owns my place, he's at 450 now but peaked near 600. At the moment he's using a big twenty milligram serving of the reta and tirz blend but wants extra. I've thought about trying cagrilintide.

Grateful for the input. My struggle with excess weight has lasted quite some time. This compound brings on tiredness along with stomach troubles, yet it activates the proper pathways well for me. It brings to mind a certain medication mix from decades back. Hunger did not merely fade away back then, it disappeared in a strange way. Meals lost every bit of appeal, so I would stare at a plate feeling complete detachment instead of simple absence of desire. The same effect shows up now with this substance. Consumption continues, though eating sits at the bottom of my priorities. Larger doses could easily make me quit food entirely.

Back then that supplement gave me the same kind of drive along with extra pep from the large containers I picked up at the big-box store. Nothing seemed able to hold me back. I can still picture the moment I turned down dessert for the first time and felt stunned by my own resolve. At the same time I was probably moving toward some kind of heart trouble, so the eventual prohibition was probably wise. Even so I ended up looking pretty sharp. My opening supply of the latest version shows up today and I cannot wait to see how it goes.

BlueThistle84archiveJun 23

The time for half of this to clear out runs about thirteen to sixteen days. Doses at three six or nine milligrams would therefore cover roughly six three or one week in turn. Pairing it with the other two would mean beginning even smaller around one milligram before stepping the amount up which stretches the supply longer still. I sure hope this one skips the awkward acidity demands that the comparable option needs.

QuietMarble20archiveJun 23
↳ replying to @WarmMeadow92

Combining these would actually let me hit my target weight. So far I have dropped twenty two percent of my starting weight with the first one and my goal is losing about forty five percent overall. Another twenty percent from the other would fit me just right.

WarmMeadow92archiveJun 23
↳ replying to @QuietMarble20

Findings from the animal trial look promising assuming they carry over to people because beginning the backup medication later seems to give more fat loss after the first one stops working well. I guess it's about time for me to pick up some of that backup if I don't have it yet.
