I'm moving over to a pen and came across the idea of using twice the usual water when reconstituting to cut down on dosing errors and leftover waste in the cartridge, which seems sensible. With 30 mg of peptide in a 3 ml vial that I plan to turn into 6 ml total liquid and split across two 3 ml pen cartridges after filtering, how should I handle it? My initial plan was to add 3 ml water to the vial, pull 1.5 ml of that mixture into a syringe and filter it into the first cartridge, then pull the rest and add the same amount of fresh water before filtering into the second cartridge. Any better approach that keeps the peptide-to-water ratio the same in both?

Testing the pen itself for accuracy should take care of any inconsistencies. Larger injection volumes can also raise the chance of injection site reactions. I've always heard the reverse: less BAC water gives more precise dosing. I normally use 2 ml BAC for most things except one peptide where I reconstitute with 2 ml, draw it into a 3 ml syringe, split it into two cartridges at 1 ml each with a fresh needle, then top each cartridge off with another 2 ml of BAC.

I was mainly thinking about the volume lost during filtering, so measuring before the filter step would let me know how much extra water to add. The accuracy point came from a video suggesting pens aren't great for very small unit counts. Ten units twice a week might be large enough that it doesn't matter. I was going to pull whatever is left in the cartridge with a syringe and have my husband use that dose so nothing goes to waste.

They probably meant under 5 or 10 units. I wouldn't dilute my peptides that heavily. My pens stay accurate down to 5 units and I haven't checked anything lower.

There's a useful write-up on using air to flush the filter and recover more solution. More BAC water means a weaker solution so any small measuring mistake has less effect on the final dose, though with pens this is less of an issue. When splitting into two cartridges my current steps to cut loss and keep things even are: draw 6 ml BAC into a 10 ml syringe, add 2 ml of it to the vial, pull the mixed solution back into the syringe with some extra air and mix gently by tilting, then filter into the first cartridge and the second, finishing with an air flush on the second one.

Which peptide needs that much dilution? First transfer the mixed solution into the empty cartridges and check that both have equal volume, then add the remaining water to top them up. I usually go no higher than 2.4 ml total split into two 1.2 ml portions and add another 1.2 ml to each cartridge. You have to purge air afterward but it avoids filling mistakes.

Focusing on vial-to-vial differences in actual peptide amount can outweigh the benefit you're trying to get from extra BAC water. I've combined two vials from the same batch into one 3 ml cartridge to even out any big over- or under-fills and because the BAC brand is hard to come by here. At the end I just draw out whatever remains in the cartridge with a syringe. Pen needles feel better to me than going through a rubber septum again, even if my skin is thick and needs more pressure.

Putting 6 ml into a 30 mg vial means injecting 80 units instead of 40 units for a 4 mg dose.

A quick way to decide total reconstitution volume is to take your planned dose per injection, divide the vial contents by that dose to get the number of injections, pick your preferred injection volume, and multiply the two. For a 6 mg dose from 30 mg that gives five injections; at 0.40 ml per injection you need 2 ml total. That yields 15 mg per ml, well under the solubility limit for this peptide.
