why am I losing two full doses of volume when filtering reconstituted tirzepatide through a small PES syringe filter

The original math checks out for six doses from 1.5 mL, but actual yield drops to four because the 4 mm PES filter absorbs roughly 7 units per pass and the syringe dead space adds more loss. Users report 10 % typical loss on small volumes and suggest starting with extra BAC water, pre-flushing the filter, and pushing air at the end to recover liquid. Larger draw volumes per dose also reduce the relative impact of filter hold-up.

I have a 30 mg vial and want 5 mg per dose. After adding 1.5 mL of BAC water I should get six doses at 25 units on a 1 mL syringe, but I only end up with four. I draw a little extra water because I filter through a 0.22 μm PES 4 mm filter, yet I still lose two full doses somewhere. Do I need to add more BAC water to account for the filtering step?

The math lines up with what should happen, so the drop to four doses is expected. The 4 mm PES filter absorbs around 7 units each time you filter. Multiply that by four doses and add the dead space left in the syringe and you can easily lose the equivalent of two doses. Using 2 mL of BAC water instead would get you closer to the six doses you were counting on.

You would need more details to pinpoint where all the liquid is going. A 13 mm filter can hold back 15 to 28 units. I use 3 mL and 5 mL syringes for the BAC water and filtering steps; those hold back a few units themselves and are less precise. I usually see about 10 % loss on small volumes. I push some BAC water through a new filter first, then when filtering the reconstituted solution I leave extra air in the syringe so I can force air through at the end to push out a little more liquid. I also draw at least 40 units per dose for these peptides and 50 units for other compounds to reduce the effect of filter losses.

That video did a good job showing exactly where the liquid disappears. I had read about the loss before but seeing it happen made it clearer. Thanks for sharing it.
