why won't my stored peptides dissolve properly during reconstitution

I stored some peptides in the fridge for a few months and they refuse to dissolve even after sitting out for an hour. The first ones from the batch worked okay. Could I be making a mistake somewhere?

I have kept some for years in the freezer without any dissolving trouble. Fridge storage should not cause problems even over long periods.

Did you bring the vial to room temperature first? It can still need up to twenty minutes to dissolve completely. Issues can happen with any source.

If the bacteriostatic water was also cold, everything needs time to reach room temperature. Once warm, shaking the vials can help the powder dissolve. Unopened vials store better in the freezer than the fridge and should be warmed only when ready to use.

I always heard shaking was bad. I tried it anyway after eight hours at room temperature and the clumps are still there.

The pH or an additive in the water could be the culprit. When two different peptides both fail to dissolve it points to the water, since each peptide needs its own pH range to go into solution.

What temperature did you keep them at? How did the powder look before adding water compared to when you first got it? Did the vials still have vacuum when you opened them?

The fridge was at thirty-eight degrees. The powder still looked white like sugar.

Try a different bottle of water and see if it dissolves the powder. Replacing the water costs less than losing the peptides.

Avoid non-pharma bacteriostatic water because it is not tested consistently and can have no alcohol or wrong pH. Only use proper pharmaceutical grade.

I agree with that view. There is no reason a peptide would be harmed by normal handling during mixing.

I have heard one compound sometimes needs a more acidic pH, while the two in question work at neutral. My last water tested at 5.4 and still dissolved fine. If crumbs remain I might filter, but only after being very sure of the product quality. People rush to use it anyway.

Shaking mainly creates foam that is annoying to work with. It would take something extreme to actually harm the molecules.

It turned out to be one of the peptides that was bad. The water must be okay since it worked on the other product. Still, the stopper on that bottle looks odd compared to others I have used. New water helped a little but not completely. Should the old water be thrown out too?

Everything turned out fine provided nobody shifted position or created any noise at all since they could simply keep going without noticing.
