I received some NAD+ but I'm unsure how to handle it. I don't know if it's buffered or what that even involves. Videos suggest 5 ml for reconstitution but dosing and schedule are unclear with lots of conflicting advice online. Any guidance would help.

Have you stayed with daily use the whole time? My ferret runs 250 mg twice a week after beginning at 50 mg three times a week.

Yes, I began at 25 mg daily then moved to 50 mg following the chart. Nothing was noticeable so I went to 75 mg and still feel no difference. I'll be adding motsc soon.

RS began with that same protocol and is now at 80 mg three times weekly. It has noticeably helped with weight loss. Will add 5AM soon. Tried Mots but had an allergic reaction.

MOTS-c appears to trigger the strongest unwanted reactions, as if the immune system is trying to quarantine the introduced molecules.

I ran NAD+ together with the full mitochondrial mix of ss-31, MOTSc, and NAD+ at 100 mg on Monday, Wednesday, Friday. I also took TMG for methyl support. General guidance suggests 1000 mg TMG daily or only on NAD+ days. No precise tests exist yet for methylation needs.

I'd like to say yes, but I'm also on reta so any energy increase could come from weight loss or the combination. I had more energy on reta plus NAD+ than on Wegovy alone.

Research shows more energy when NAD+ is paired with Tirz. It made a big difference after feeling drained on Tirz by itself.

Electrolytes helped a lot with tirz side effects for me. I stayed at 25-50 mg three times a week and it raised my energy baseline even if it didn't fix anhedonia. Under 40 the natural systems may not be as depleted so effects could be milder. It still stings even when buffered. I reconstitute 250 mg vials with 2.5 ml and inject slowly. Effectiveness drops after about four weeks so I avoid larger vials.

No one has addressed the buffering question yet. Older NAD+ was very acidic and caused bad reactions. Buffered versions were meant to fix the pH but many still sting. A good target range is around 5.0 to 6.0 for comfort while keeping stability. I have tested and adjusted every batch I've received. pH strips are easy to get.

I've never had injection issues with any buffered NAD+ from several sources. Zero site reactions across the board.

From earlier posts it didn't help much subq so you switched to IM? Do you prefer oral versions now? I just started today at 25 mg and it stung a bit even though labeled buffered. I used 3 ml for a 500 mg vial and may add more or try IM instead.

Injecting into the muscle has produced superior outcomes for me over doing it under the skin. The advantages become more apparent the more time passes while using this method. My routine involves 50 milligrams each day. It never seemed logical to me, considering what I understand about NAD, to administer it just one or two times weekly or according to the mitochondrial restoration approach frequently discussed. Experts studying the NMN and NR precursors strongly recommend daily intake over several months before experiencing complete effects. I fail to understand how NAD+ shots would differ in that regard. The oral versions of NMN and NR also failed to provide noticeable results for me. Additionally, I have consistently mixed my 500mg NAD using 3ml of the solution.

I was mistaken earlier. I usually reconstitute 500 mg of NAD+ with 5 ml of hospira, which may be reducing the site reactions.

Dosing varies a lot. Without knowing age or history a conservative titration is safest. I started at 100 mg daily and stayed there because I'm older and had used precursors for years. Even buffered versions sting. I got used to it and stopped trying to adjust pH. So far I haven't noticed much benefit and wonder if higher doses or switching to oral would help.

Did you work out after the NAD+? I never viewed it as a direct fat-loss compound. It felt more like creatine where you only notice it when you train. My reps increased and I could progress weight faster. I'm looking for current promotions to run the protocol again before summer.

Further reading suggests MOTS-c may have been the main driver. Still, running the full protocol for synergy seems worthwhile. MOTS-c shows stronger effects on reps, fatigue delay, and lactate threshold while NAD+ helps more with recovery and daily energy.

I injected three times a week, usually not close to workouts. I felt nothing at all, same as with creatine which also did nothing except raise liver enzymes. It was sold as something that would increase fat burning and energy but that didn't happen for me.

How much creatine were you using? I stick to 5 mg a day and sometimes miss doses. How old are you? Most mitochondrial support is aimed at older people. Sleep is still the biggest factor for energy. If sleep is good but energy is low, bloodwork can reveal deficiencies like low iron or B12.

I was taking 5 mg of creatine daily and I'm almost 48. Bloodwork is fine except when creatine is involved. NAD+ simply did nothing for me. I've looked into the other items mentioned but haven't started any yet.

I agree that benefits beyond anecdotes are limited. My main reason was recovering energy after a very restrictive diet. Older users with more accumulated stress are more likely to notice a difference, similar to B12 for someone deficient. I'll probably run another cycle in a few months but don't expect GLP-1 level changes.

I reconstituted 1000 mg of NAD+ with 4 ml bac water, filtered it, and ended up with 5 ml total. Does NAD+ add volume after mixing? The solution is noticeably thicker than my other peptide mixes.
