Just wondering how to handle higher concentration peptides per vial. The usual approach is to add 1-2 ml of bac water per 10 mg of compound. That works fine with 10 mg or 20 mg vials, for example 1 ml into a 10 mg vial so a 2 mg dose is 20 units on a 1 ml syringe. With a 40 mg vial though, even adding the full 3 ml means a 10 mg dose comes to 60 units, which feels like a lot. At the same time the ratio drops below 1 ml per 10 mg. Two questions come up: what is the maximum volume that can be injected subcutaneously at one time, and is it a problem to reconstitute with less than 1 ml of bac water per 10 mg?

A reverse calculator can help figure out the exact amount of bac water needed based on the target draw. I usually add 2-3 ml of bac water unless the peptide tends to sting, in which case I dilute more. There are also cases like very small doses where different volumes make sense. For ordinary peptides I try to land around 20 units per dose for easier measuring. Insulin syringes top out at 30, 50, or 100 units, and those volumes all appear acceptable for subq use.

The idea of one common rule for all peptides is not accurate, and the amount really varies by compound. I prefer to keep injections at 35 units or less and at least 10 units when possible. Some peptides are too reactive for that approach, but I avoid most of them. In the past I reconstituted one compound very weakly at 12 ml for 70 mg, yet a later batch worked fine at 3 ml for 50 mg. For another peptide I combine three vials with 2.25 ml total and filter into one cartridge to hit 10 units per 1 mg daily dose over a cycle. I keep tirz at 22 mg per ml simply because that is the strength I began with and have continued using.

So another question is whether concentration itself matters. Is it safe to reconstitute 40 mg of that peptide in 2 ml of bac water?

You could add far more bac water than needed and still be fine. There is no need to worry about being too dilute. For the two peptides mentioned, 20 mg per ml works well based on prior experience with one of them, and the other should behave the same way.

I'm moving to the next vial size. I'll add 2 ml bac water so a 7 mg dose comes out to roughly 0.23 ml. That should give just over eight doses per vial. Using 2 ml now that the per-injection amounts are higher feels better than the previous 7-8 mg shots and also makes drawing easier. I'll apply the same approach to the 30 mg vials.

Alright, got it. So concentration is not really an issue except with certain stingy peptides, and then it is only about comfort during injection.

I reconstitute, transfer into a larger vial, and then add more bac water. For example with a 3 ml vial holding 50 mg I start with 2.5 ml, move everything into a 5 ml sterile vial, and add another 2.5 ml for a final 5 ml total.

It really depends on how much bac water ends up being injected and how much dilution is wanted. For some reactive peptides extra dilution helps. I chose 2 ml for one that tends to sting, though the injection site also plays a role.

I am not sure concentration should be viewed only as a comfort factor. For the specific case of 40 mg in 2 ml that is clearly fine, since one approved product comes at 30 mg per ml. That sets a precedent that 20 mg per ml is acceptable. The open question is how high is too high, and it may differ by peptide. Reconstituting the same 40 mg vial in only 0.5 ml would reach 80 mg per ml, or 120 mg per ml with a larger vial, and it is unclear whether those levels are advisable. Some peptides are more likely to aggregate at higher strengths, so pushing the upper limit may not be wise with those.

Actually the example was about the other peptide, but the direction is the same. I am wondering if there are any recommended concentration patterns for specific peptides and whether the final concentration changes stability, shelf life, or effectiveness. I have not found research on that point, which is why the thread started.

One thing not yet mentioned is that higher concentration means the syringe must be loaded more precisely, since small volume differences create larger dose variations.

For research use where the goal is simply to avoid issues later, I would choose more bac water rather than less. Extra volume gives more room for the peptide and can help maintain the preservative effect.

The exact concentration used in the trials has not been publicly released. Since the compound is not approved, full formulation details remain unavailable. I agree with the suggestion to use more bac water rather than less.
