I'm struggling to get a reconstituted vial of this peptide into a pen cartridge through a filter. Near the end it just refuses to move through at all. I even tried a low-gauge luer needle. Any idea why it's happening or what to try next? I really wanted to filter because I skipped wiping the vial top with alcohol. Has anyone else seen this? Maybe it's just this peptide?

This happens to me with those 4 mm filters pretty often. Grab a fresh one. Some of them just seem stubborn. I wet mine first with bac water and that sometimes makes a difference, though not every time.

Thanks. I checked with chat and it said this peptide doesn't filter well because it sticks and clogs right away. So I'm just going to skip the filter on this one.

Which filters have you had the best luck with and seem reliable? I've been considering them but heard some brands clog a lot, up to a quarter of the time. The higher-end ones can run pretty expensive per piece too.

One suggestion is to use PBS as the buffer for this peptide, while others recommend a small amount of sodium bicarb instead because it makes filtering easier.

Not sure about this one, but with reta and mots I had similar issues. Adding an extra needle into the cartridge for venting helped. Did you try that?

The vent needle really matters. Without it you can end up forcing the stopper right out of the cartridge. I found that out the hard way.

No experience with this peptide, but I tried filtering some other compound vials with a 4 mm filter and it wouldn't move either. Ended up switching to a 13 mm filter instead.

13 mm looks like the smallest size people usually recommend, but I need to check further. A quick search on syringe filter types and loss rate turned up several useful links. I might go with a good 13 mm once I find one, but the whole point was filtering for endotoxins and it seems they can't handle that.

I checked with an AI and it looks like the 4 mm size is actually the right choice for peptides. The membrane material matters a lot for keeping loss low, and PES or PVDF are preferred because they bind less. Filter diameter also changes how much volume stays behind, with smaller ones losing far less than bigger ones. For volumes up to a few milliliters the 4 mm is best, while 13 mm suits larger amounts.

Videos on filtering usually show the 4 mm size being used for small batches like one to four 3 ml cartridges. They also sell the gear including those filters. When you're only doing one vial the bigger size means noticeable loss. Checking steps from other sources under their filtering section is useful too.

I went ahead and ordered the 4 mm PES 0.22 ones. Twenty-five of them came to thirty-two dollars. Plenty of comments say results are inconsistent, but that seems to be the cost of trying to filter.

Wetting is for filtering liquid, not for filtering air.
