When reconstituting my peptide the bacteriostatic water did not get drawn in the usual way so I had to push it. After adding one milliliter the plunger was forced back outward. What might cause the vial to show positive pressure? The liquid stayed clear and mixed without trouble.

Is this the first vial from the kit or have the others also lacked vacuum?

I have come across a few vials without vacuum. A vent needle may be needed in those cases.

This is not entirely uncommon. Since it dissolved clear I would consider it acceptable.

If I am reading the description correctly the pressure may have come from the way the water was added. Air could have been pushed in after the initial pressure was applied. Cloudiness would be more concerning than pressure. It is also possible the peptide was agitated enough to create gas. I always vent first with a second needle and let the water run gently down the side rather than letting the vial draw it in. Only a light swirl afterward is used.

I have never encountered one without vacuum and would worry the seal had been compromised. Was the batch checked for sterility and endotoxins?

Not every kit arrives with vacuum. I have seen some without it. They were not tested but performed as expected with no problems.

No vacuum makes me picture someone simply filling and crimping vials by hand. That seems questionable.

It came from a kit and the other vials from the same kit have been normal. I will watch for any cloudiness. I have always let the water be drawn in and directed it down the side. The vials are then placed on a roller to mix. Recent kits seem to have inconsistent pressurization and the caps are difficult to remove without damaging the seal.

I have had a couple without vacuum and testing came back fine including endotoxins. That is probably how they are filled. How long have you been ordering from that source?

Thanks, I was just wondering. I have never ordered from them. The one I have that someone else had trouble with is still clear. I plan to wait until the end of the month before using it.

It is surprising that no one here seems to grasp that missing vacuum indicates a failed seal and lost sterility. It also allows oxygen and moisture to reach the peptide and cause degradation. Vacuum is part of the normal lyophilization process and its absence means something went wrong. Clarity or subjective feel does not confirm safety or potency.

Do not try to pull the cap straight off in one motion. Lift one edge until it clicks then open from the opposite side. That prevents the metal from peeling away.

The vial had no vacuum which sometimes happens. Adding the bacteriostatic water created positive pressure because the liquid took up space and the air could not escape without a vent.
