How soon did you start feeling effects after first dose?

33 replies29 peopleApr 10, 2026☆ Follow
Summary

How quickly do people notice appetite changes or side effects after their very first dose of tirzepatide or similar peptides?

Most users report noticeable appetite suppression or fullness within a few hours to 24 hours of the first injection, though a minority feel nothing until the next day or later. Timing and strength vary widely even at the same low starting dose, with some describing instant food-noise reduction and others needing multiple doses before consistent effects appear. Side effects like nausea or digestive issues also show up at different points for different people.

Not sure whether this is just in my head or if I react fast, but less than a day after my first 2 mg shot I already have zero interest in food. I started low because my BMI is on the healthier side, yet I genuinely feel full even though I barely ate. Never tried anything like this before so I have nothing to compare it to. Wondering if others felt something this soon or if I'm imagining it.

LevelThistle44archiveApr 10

That lines up with normal timing for most people. I figured the peak would hit around 24 hours and was wondering the same thing when I began, whether anything was happening maybe half a day in.

CopperBeacon48archiveApr 10

I noticed changes the very next day even though I went into the first shot feeling pessimistic. Suddenly I was eating clean with no cravings for sweets, while my dad needed two or three weeks before the food noise faded. It's odd how differently two people can react to the exact same batch.

SteadyLedger26archiveApr 10

I can usually tell something is happening about a day after dosing, and it reaches its strongest point by the second day.

SharpTimber40archiveApr 10

I notice it within an hour or two. Stomach injections seem stronger. You will probably feel it clearly the following day, and once it builds up the sensation stays with you even on days you would normally dose again.

LevelBramble55archiveApr 10

The day after my first shot my whole body ached. I have not had any side effects since that initial dose.

AmberCompass54archiveApr 10

I felt something within three to four hours.

PlainQuill53archiveApr 10

With this medication I can predict exactly when the bad stomach issues will hit, usually 36 hours after the first shot following a break or dose increase. Appetite suppression, though, shows up by the time I wake up if I inject right before bed.

CopperSignal27archiveApr 11

I keep noticing that people who start these medications at low doses while not carrying a lot of extra weight often report fast or strong responses. It makes me wonder if that pattern is more common when someone has less weight to lose and fewer metabolic issues. After dropping a lot of weight I still needed to push tirzepatide up to 15 mg over a month before hunger really quieted, and I used small frequent doses to limit side effects.

LevelWillow41archiveApr 11

For me the first signs appear around the three-hour point.

Thanks for the info. I have been let down that I am not getting the same dramatic results others describe. I am still early but so far appetite and everything else feel unchanged.

RustCompass29archiveApr 11

The day after my shot every bite felt like it landed in a heavy lump. I got full faster and cared less about food overall, which was nice since I normally deal with constant food thoughts.

PatientMeadow16archiveApr 11

I noticed it the next day during my lunch break at work. I could only finish half my usual meal before feeling full. It was a strange mix of disappointment and confusion at first, realizing I had been overeating for years. After three and a half months I am used to smaller portions and sometimes have to remind myself to eat enough protein. No need to rush higher doses.

CopperSignal27archiveApr 12

When I began semaglutide after already losing a lot of weight, the first clear sign was feeling too full to finish even a modest plate, something I do not think I had experienced before.

Took my first 2 mg dose and felt nothing at first, even wondered if it was inactive. By day two or three I noticed a shift, not exactly full or hungry, just neutral toward food and able to stop when I had enough. The next two mornings I woke at 4 a.m. with headaches despite staying hydrated and keeping electrolytes up. It seems to be working, just not the way I pictured.

PlainMeadow55archiveApr 13

I began a low dose for the first time last week and felt something within half an hour. It was like the constant background noise about food suddenly disappeared. Sticking to my calorie target became oddly easy the rest of the day. I picked up a planned snack and left half of it, which never happens, and approached dinner the same way. With the low dose the effect faded after a few days and hunger returned, so I am looking forward to going higher eventually.

I avoid stomach injections because they hurt more. I use the outer thigh instead and notice the effect just as fast while staying more comfortable.

SlowSparrow81archiveApr 13

After the first shot I would have sworn it worked instantly. I injected an hour and a half before my usual breakfast time and did not eat until 11 a.m. because I had no hunger. The second shot took until the next day for the full feeling to appear. Third shot is due Wednesday so we will see.

SlowThistle36archiveApr 16

I have stayed on 2.5 mg for a month. When I inject around midday I notice reduced hunger signals by late afternoon or early evening, but the clearest effect always shows up the next morning. That window from the first few hours through the first full day is when I feel it most, and it keeps getting stronger as doses accumulate.

WarmHarbour28archiveApr 17

I began more than a year ago at 2.5 mg and could feel it within minutes. After thinking about food for decades the sensation is strange. At my current higher dose it takes a little longer to appear. One of the early higher doses gave me a brief buzz that I still cannot explain.

It took about a day for me. This was also my first time trying any peptide. I do not know if I am a fast responder, but after nine weeks I have dropped 23 pounds from 207 to 184. I am a 5'7" female with an athletic build and probably have another 20 pounds to go. I just took my second 7.5 mg dose after skipping one week of the 2.5 mg early on.

SlowSparrow81archiveApr 18

I felt my first 2.5 mg dose within an hour, which felt odd. By the third and fourth shots it took several hours, with the full effect arriving the next day. Now on the fourth day after the fourth shot I felt stomach rumbling this morning for the first time since starting, and yesterday afternoon and evening I was a bit hungrier too. I had thought I was a fast responder, and my starting BMI was only 26.5, now down to 25.

BlueAlder28archiveApr 18

My first three shots at 3 mg each produced a reaction around 18 to 20 hours later. It felt like a gag reflex where I stood at the sink spitting every ten seconds because I could not swallow the extra saliva. I nearly vomited the first time. The next two were more like an upset, knotted stomach. After that the reaction disappeared, so I now time later shots for days when I can manage any issues during work hours.

AmberMarble84archiveApr 18

Same here, noticeable 12 to 24 hours after the first injection.

CopperAnchor42archiveApr 25

I weighed 358 pounds when I started in February at 2.5 mg. I injected right before bed and woke up having to force myself to eat that day and the next. The effect felt strong for more than 48 hours and lingered somewhat for six days. That intensity lasted the first few weeks. Now at 7.5 mg after losing about 25 pounds I do not feel it until day three or four and it only lasts a day or two.

BlueKettle13archiveApr 25

I took my first dose at 7 a.m. I normally skip breakfast, eat lunch, snack through the afternoon, and have dinner. By 8 p.m. I realized I had not touched food all day and had no desire to. I responded strongly to tirzepatide. Six months later I switched to the other medication but still do not think I respond to it at the low dose I am using. Time will tell whether I go back.

GreenBeacon77archiveApr 25

It hit me very fast, within a couple of hours. The next day I could barely eat. I ended up using meal-replacement shakes for lunch during the first month just to get some balanced calories in, and dinner portions stayed small too, all on a 2.5 mg dose that I kept for months.

SlowLedger41archiveApr 25
↳ replying to @GreenBeacon77

I have also been using protein shakes to get nutrients in. It is surprising how little I think about food now. I used to snack without even realizing it.

SlowLedger41archiveApr 25
↳ replying to @BlueKettle13

I thought about trying the other medication but read it can raise heart rate, which worried me a bit. What prompted you to switch from this one in the first place?

BlueSignal76archiveApr 25

I began with the branded version. I noticed an effect after a few hours, but during the first week it only lasted three or four days. After more than ten weeks the same 3.5 mg dose now lasts the full week.

SharpCinder28archiveApr 25
↳ replying to @SlowLedger41

On the upside, a higher heart rate can sometimes come from lower blood pressure, and the other medication may improve LDL more than this one. There are also ways to combine them or use alternatives for stacking, though one option is cleaner for that purpose. I will always prefer this medication and still have more of it stocked than the other.

BlueKettle13archiveApr 26
↳ replying to @SlowLedger41

The main reason was the extra fat-burning effect, but honestly it has been only average for me. It works about the same as the first medication with a bit more heartburn. My resting heart rate rose roughly ten beats per minute, from the 70s into the 80s, while my workout max stayed at 160 and never went higher.

WarmQuill45archiveApr 26

My experience was unusual. About an hour after the first shot I went to lunch and ordered a normal meal. Halfway through I suddenly felt strong nausea and could not imagine taking another bite. That exact reaction has not returned even though my appetite is lower overall, and I ate the same meal weeks later without issues. It might have been first exposure, an accidental capillary injection, or even placebo, but it startled me at the time.

QuietKettle56archiveApr 26

I have been on it for six months. It may sound odd, but on day two I first noticed I had zero interest in snacks and did not want dinner either. Nearly six months later I still can barely eat. I make sure to get protein and nutrient shakes and occasionally manage small meals, though I am trying to improve that habit.
