MOTS-C not working?

12 replies10 peopleApr 22, 2026☆ Follow
Summary

Why am I not feeling effects from MOTS-C and what dosages or protocols do others use?

Users report mixed results with MOTS-C at doses from 2.5 mg to 8.5 mg several times weekly. Some notice improved workout output or cardio energy while others feel nothing or even fatigue. Several suggest running SS-31 first to support mitochondria before or alongside MOTS-C, especially for those over 40. A few who saw no subjective change still recorded objective increases in training volume.

What this discussion establishesWhere people disagree

Whether lack of noticeable effects means mitochondria are already optimal or simply not ready for MOTS-C without prior SS-31 support

Still open

Optimal sequencing and whether MOTS-C and SS-31 can or should be run together for best results

Nothing here is advice.

Is anyone else taking MOTS-C and not noticing any improvements? I've worked up to 8.5 mg before training four or five days a week yet still feel nothing different. What amounts are others using?

Same here, no noticeable change at all. From what I understand MOTS-C needs working mitochondria because it only activates the AMPK pathway. We might have an issue there. Many recommend fixing the cells first with SS-31 and maybe FOX04-DRI to clear senescent cells. I'm planning to try that approach.

I can only tolerate 4 mg. Anything at 5 mg feels like too much. Maybe those who don't notice anything already have mitochondria that are functioning well. They could also be younger and haven't yet run into the age-related drop in metabolism. If you're past 50 and feel no difference then you're fortunate.

I should have said I'm 43 so based on what I've seen I expected to notice something since mitochondrial decline has probably begun. I might stop, get an SS-31 kit and try that first.

That's the usual approach but newer studies indicate they can also be used at the same time. I did SS-31 first. NAD levels also begin to drop around 40 so you might still have solid natural mitochondrial function. At 67 mine has declined so the effect was clear for me.

I use 7 mg every other day. It helps with cardio and gives clean energy. I did a month of SS-31 back in November. It left me completely drained but seems necessary. I'm getting ready for another round of SS-31.

I thought about trying SS-31 before MOTS but decided to skip it and just train five times a week plus run twice a week so I may not be the best candidate anyway. I started directly with 5 mg of MOTS twice a week before runs and it works well for me, keeping tiredness way down.

I'm using 5 mg of MOTS-C three times a week. On week four I really like it and can feel a clear difference while training.

I just began 2.5 mg three times a week last week and I think I'm already noticing it on my longer run days of seven miles or more.

Same experience, finished one vial and felt nothing at all. I still have nine more vials stored away. I might try again later. Same thing happened with GHK-CU, one vial and zero benefits noticed. I'm not usually affected by placebo. Tirzepatide on the other hand gave clear results that showed up on the scale.

Looking back at my training log I see I've been adding a couple extra exercises which adds about fifteen minutes per session so my total output has probably gone up around twenty-five percent even though I don't get any energy rush feeling.

Interesting that it's producing changes you don't feel subjectively. I'm looking forward to trying it myself especially for cardio performance.

WarmWillow50archiveApr 27

The only change I noticed from MOTS-C was feeling more tired. I don't believe my mitochondria were ready for it to work properly. I stopped, focused on mitochondrial support, plan to do SS-31 next and then try MOTS-C again.
