Roche's 48-week phase 2 data show solid weight-loss numbers along with lower dropout and better tolerability than today's GLP-1 medicines. The dual agonist, once called CT388-103 and now Enicepatide, produced 22.5% weight loss at 48 weeks without any plateau, 26.1% of people lost 30% or more, and 73% of those with prediabetes returned to normal blood glucose. At the 24 mg dose the dropout rate was only 5.9%, roughly half that of other drugs in the class.

Very similar to tirzepatide in many respects except it is a biased agonist toward cAMP at both GLP-1 and GIP, while tirzepatide is biased only at GLP-1. Weight-loss numbers look comparable and the low dropout rate stands out. I could not locate the actual paper to check side-effect rates. Weight loss had not plateaued at 48 weeks, so longer treatment should produce even more loss. It is difficult to predict how any of the new non-triple GLP drugs will fare; retatrutide is simply more effective and other triple agonists in development will likely be too. This candidate might stand out through fewer side effects given the low dropout, which could help it in the market versus tirzepatide. I see no realistic path for injectable GLP-1-only drugs in development unless they are made cheap and launched quickly before many more options appear.

With multiple approaches now available for extending duration, I expect the first compounds will be close matches to current ones but tweaked just enough to skirt existing legal protections. There could also be attempts at versions aimed at changes in people who respond poorly, though making those will likely prove tough. On top of that, some patients may resist anything but the original versions, much like what occurred in heart medicine.

These days you might need to search around for the newest developments. To me the truly game changing material still depends on that biological situation at the firm. No reason to let go now if its worth could multiply ten times overnight Haha

Actual rivals around here might reduce overall costs. I'll keep getting my supplies the usual way, though this shift could transform things for anyone avoiding unofficial channels.
