As someone fairly deep into nootropics I am surprised Selank and Semax have returned to popularity. They strike me as average within the broader nootropic space. They can work but plenty of stronger options exist if they suit you.

I work full time as a software engineer. Ranking my nootropics by effectiveness, the top spot goes to sub-hallucinogenic doses of pharmahuasca using 5-meo-dmt with an MAOI. The risks, sourcing hurdles for most people, and required diet changes make it hard to recommend. Next come creatine and high-dose EPA fish oil. I keep a steady 6G+ total Omega3 intake daily in liquid form. For creatine I use the liposomal version twice a day with good hydration. If you are not already doing these basics then focus there first. Other mentions include Tropisetron, Mag-L-Threonate, BPN-14770, Clomipramine, JNJ-42165279 and Tak-653.

Just to clarify up front, I am asking this out of genuine curiosity with no negative tone. Your first post seemed to say Semax and Selank are mediocre and that better options exist for similar benefits. The follow-up post about your favorites did not clearly connect back to replacing Semax or Selank. Do you have any nootropic suggestions that would directly outperform them in your experience?
Just adding my voice here to back that query, supposing this isn't meant for whoever replied first.
Also worth noting is how one of those peptides clouds my thinking while the other one does a great job quieting down mental distractions. More options like that would be nice if they avoid adding much energy or alertness. The ones mentioned earlier definitely don't fit the bill.

DMT at sub hallucinogenic doses taken orally modulates the 5-ht2a site. That is strongly pro-cognitive and the MAOI keeps the molecule from breaking down. I could not work on high dose DMT. Since that may be too far for most people, a solid alternative to Selank is NSI-189 taken with Magnesium L-Threonate. Some compounds now in development for fragile x look interesting, especially zatomilast which seems to boost dendritic density. No RCTs exist yet in healthy humans so I hesitate to endorse, though caregiver ratings and coding scores are positive. If you want to go further there is nightly vTNS. I am also using a fair amount of tropisetron lately.

I wrote a very lengthy reply and it got rejected by the anti-spam filter then disappeared. I will keep this short. Selank's exact mechanism is not fully known but it ultimately increases BDNF. That is not always beneficial. Upregulating nrf2 and trkb pathways to raise endogenous glutathione seems better for BDNF. For noise I prefer guanfacine and l-theanine over many anxiolytic RCs I have tried. Guanfacine is remarkably pro-cognitive. For a direct swap in place of Selank try NSI-189, now called ALTO-100 or similar. I am not as enthusiastic about it now but brain scans showed it repairs and increases hippocampal volume in both healthy and depressed volunteers. When I used it I felt very sharp and not depressed. It is currently in phase 2 trials.

This compound has really helped me relax and sharpen my attention throughout the day. Using it right after reaching the office brings me outstanding personal effects. It seems to reduce my tendency toward excessive rumination. Making the nasal version at home works fine as well. Excellent video guides explain the steps online. Only the dry form plus some salt solution and a dispenser are needed. It surprises me that doctors do not recommend it instead of their usual tablets.

I had success using nasal versions of two compounds against the mental cloudiness that stuck around after COVID when things stayed fairly light. The usual approach calls for muscle shots if that option fits what someone can handle. Research is also exploring a certain peptide paired with a related molecule for similar effects, though fewer papers exist and nothing has involved human subjects yet.

Anyway it's alright. This isn't purely a peptide though it works more like a blend containing enzymes peptides and amino acids. The material comes from pig brains after enzyme processing and includes several nerve growth factors such as the brain derived the glial cell line derived the nerve growth and the ciliary ones plus unbound amino acids together with different small peptides.

For the past fourteen days I've been injecting this compound under the skin daily but I notice zero changes even though I'm using a substantial amount each time. There's simply no effect at all. I'm planning to switch over to the version that goes in the nose instead. If someone has a preferred way to prepare it or a good mix with salt water I'd appreciate hearing about their experiences with that. I should mention I also take another medication prescribed by my doctor which works really well for me but I have to watch the amount carefully because exceeding my limit leads to strange mood drops in the afternoon. I thought this new one might provide some benefit but up to now it hasn't done a single thing. Perhaps using it nasally will make a difference.

I looked over an online calculator that lets you input the desired concentration for each nasal spray and then works out the proper blending amounts. Registration was necessary to access it, but the tool proved quite handy for handling dosage and reconstitution figures right away.

It works that way sometimes. Little problems would annoy me and I'd focus too much on each one. The steady calm lets me concentrate without wanting to handle several tasks simultaneously. This has helped me become kinder toward unpleasant people.

In my experience this works better when taken through the nose. The results tend to be mild no matter how it's used. My homemade nasal version provides five hundred micrograms with each spray to reach one milligram and I used the unmodified form. Nasal delivery worked better for me due to greater absorption compared to under the skin shots that go through the liver first.

I've switched to intranasal for several peps like Semax, Selank, P21, DSIP and Epitalon because I got tired of daily injections. I reconstitute using BAC saline and these spray bottles that deliver .12 mL per pump. One thing to remember is avoiding a hard sniff when dosing so the peptide reaches the upper nasal cavity instead of running down the throat, especially for the nootropic ones aimed at the brain. Tilting the head down helps keep it where it needs to go.
I just draw mine out with a syringe. I use sterile saline instead of bac water and end up putting the mix into a sterile amber nasal-spray bottle, but otherwise I follow the same filtering steps as when I pin peptides. If there's a bottle opener tool then maybe I wasted some syringes. Curious how others reconstitute for intranasal use. I've also started using my BPC157/TB500 combo vials intranasally after the Selank dose and it seems to be helping with some chronic sinus problems I've had. Might deserve its own thread, but I'd like to hear if anyone else has tried intranasal BPC or TB.

My original post on the intranasal reconstitution process is in another thread. I used the same steps for Epitalon, Semax and Selank plus the NA versions. I also deal with ongoing sinus issues. BPC and TB are large molecules so they probably won't cross the BBB easily, but they should still help topically with sinus inflammation, so you might be onto something. TA-1 and KPV have been used for that too and I made a combo spray with them, though they target viral issues more than the usual bacterial sinus infections and the combo didn't do much for my last one.
I have a question. I received my semax already in a nasal spray and it's finished. Is it best to use a fresh spray bottle for each new portion of semax, or is there a reliable way to clean the old bottle thoroughly? How long can reconstituted semax stay good in the fridge when mixed with NaCl?

You can clean the bottle and sprayer with alcohol or hydrogen peroxide. I fill it halfway, spray a few times through the mechanism, let it sit ten minutes, then rinse with saline and let it dry. You could go further with UV or autoclaving if you wanted, but I've never had a problem. My sprays have stayed clear for thirty days refrigerated and I'm still fine.

I create a nasal spray with twenty milligrams of that peptide and take one milligram per day over twenty days usually upon waking until depleted. I follow a similar pattern with the other peptide except I have tried thirty milligrams to cover thirty days and I keep the modified version in that size. Following completion I pause for a minimum of two weeks although extending it would be preferable. The first peptide gets cycled on a four month interval. Separate delivery devices hold each one. Despite working together well I separate their applications by thirty to sixty minutes. A new peptide is arriving soon featuring an adamantane addition to enhance uptake. Nasal delivery fits it well due to the compact dimensions. Typical amounts fall between five hundred micrograms and two milligrams so I begin at the lower level. Its effects accumulate thus I advance slowly and carefully continuing four to six weeks. No research on humans has been done yet but a particular paper seemed noteworthy.

I experimented with different versions of those substances. A couple of them produced no noticeable effects for me at all. One stands out as completely distinct though. I've used that one over the past five months and it's made a huge difference in my life. Now I can just relax inside my thoughts without doing anything else. There's no urge to occupy every second or to tackle issues before they arise.
