Reta users - anyone experience increase in appetite initially?

Summary

Does retatrutide cause increased appetite at low starting doses and how does it compare to other GLP-1 drugs?

Some users report a temporary rise in hunger when beginning retatrutide at low doses like 0.5-1 mg, which often improves after titration to 3-5 mg twice weekly. One person switched from semaglutide and needed higher retatrutide amounts to match prior appetite control. A receptor activation chart was shared to explain weaker GLP-1 effects but was later noted as in-vitro data that may not translate directly to real-world results. Tirzepatide was highlighted as outperforming semaglutide in human trials despite the chart.

Whether the in-vitro receptor chart explains real appetite differences or is irrelevant to human effects

Nothing here is advice.

My wife began Reta about two weeks ago using half a milligram twice a week. She has noticed times when her appetite is a little higher and she ends up eating more. Does this happen for others who start at a low dose?

Thanks for the details. I will discuss increasing her dose with my wife.

Is she gaining weight? If not then extra appetite may not be an issue. Most people feel hungrier on reta than on other GLP drugs. The chart does not apply to actual effects in the body so it does not explain the experience.

Does the chart show relative effects? I would like to see it.

The source is a 2024 peer-reviewed paper by Eli Lilly scientists. In the table retatrutide shows the best GIP and glucagon activity while semaglutide leads on GLP-1. To match one milligram of semaglutide on GLP-1 you would need roughly zero point four five milligrams of retatrutide or nine milligrams of retatrutide compared with one milligram of tirzepatide.

The chart title says in vitro assay so the numbers are from test-tube experiments. Those results do not necessarily match how the drug behaves inside a real person because the body is far more complex.

You are right I missed that detail. It shows why I am not a medicinal chemist. I will take the information with some caution from now on.

The chart would suggest tirzepatide performs worst yet a direct human study showed it produced forty seven percent more weight loss than semaglutide over seventy two weeks and better A1c reductions. I am currently using semaglutide for maintenance after stocking it as backup and my kidney labs are improving so I may stay on it longer for that non-scale benefit.
