Mazdutide 48-week study at doses up to 16 mg and user experiences

19 replies5 peopleNov 7, 2025☆ Follow
Summary

How does mazdutide perform for weight loss at high doses compared to retatrutide, and what are real-user dosing and side-effect experiences?

A 48-week phase 2 trial showed the 16 mg dose of mazdutide produced about 22 % weight loss, approaching retatrutide results, though nausea occurred in 60 % of those on the highest dose. Users discuss daily micro-dosing to reduce side effects, stacking with tirzepatide, and personal liver-enzyme changes that led one person to pause retatrutide. Opinions differ on whether daily injections are worth the effort versus weekly protocols and on how much glucagon agonism adds beyond weight loss alone.

What this discussion establishesWhere people disagree

Whether daily dosing meaningfully reduces side effects compared with weekly schedules, and how accurately short-term trial curves can be projected to 48 weeks.

Still open

Long-term solo mazdutide results and whether daily micro-dosing is superior to standard weekly regimens.

Nothing here is advice.

A 48-week trial tested several doses of mazdutide. Participants on 16 mg lost a little more than twice the weight of those on 6 mg, though the 6 mg group spent most of the study on 3 mg and only reached 6 mg near the end. Nausea hit 60 % of people at the top dose. Average loss at 16 mg reached 22 % of starting body weight, close to the 24 % seen with the highest retatrutide dose over a similar period.

Thanks for sharing these. Plenty of people are focused on reta right now, but other compounds also deliver solid results.

Reta works for many but causes issues for others, just like semaglutide and tirzepatide have for some. I am currently using mazdutide myself, though it is still too early for me to say how it is going.

Appreciate the information. Keep us updated on how the mazdutide goes. What dose are you using at the moment?

My schedule is more involved than most would want. I track daily levels with a spreadsheet that estimates how much remains in the system. The right column shows the amount present; the left shows the daily injection. I round off the hundredths and aim to increase the level by 0.2 each day. The calculator assumes a 10-day half-life. Bold numbers are future planned doses.

Are you running mazdutide by itself? Most reports involve stacking it with survodutide, so there is not much solo feedback yet. I would be interested to hear how it goes if you are not combining them.

Why inject every day? That sounds very precise.

I also take tirzepatide at 1.1 mg daily. I stay at that level because higher amounts leave me fatigued and cause stomach issues.

These drugs were never studied with daily dosing, which is one reason not to use the newer ones that way. I dose daily so my system can adjust gradually and to avoid big swings in levels. Weekly schedules mainly exist because most people prefer fewer injections. Whether daily use actually lowers side effects would need proper trials.

You are probably right that weekly dosing is mainly for trial convenience. I split reta and cagrilintide into twice-weekly shots and have avoided side effects that way. Weekly dosing leaves me with returning food noise by days five and six.

No side effects at all? That is excellent.

Tirzepatide gave me sulfur burps, loose stools, and early sleep trouble, though nothing severe. I also get the vivid, sometimes unsettling dreams that some people report with retatrutide.

One day someone will share results from mazdutide used alone.

A chart compared earlier trials by percent weight loss, but the time frames differed. I added lines showing where the curves might land if extended to the same 48-week mark used for retatrutide. Mazdutide came close, and the cagrilintide-semaglutide combination even closer.

Matching retatrutide-level results with mazdutide seems to require 10-16 mg doses according to the available studies. Those trials did not test the two drugs head-to-head, but a recent 48-week study used 10 mg and 16 mg of mazdutide.

I am not sure which study you mean. The 6 mg arm looked strong through 24 weeks. Losses usually slow after that, so extending the lines may not be accurate. Do you think mazdutide affects the liver differently?

Any weight-loss drug tends to lower liver fat, but those that also activate glucagon receptors do so more strongly. Mazdutide and survodutide are partial glucagon agonists, as is retatrutide. I have not looked into glutathione.

I thought you had higher ALT readings and switched because of that. Maybe I am mixing things up.

CopperLedger37archiveNov 9, 2025

ALT reached ten times the upper limit. I waited for a hepatologist before considering any liver support. We stopped both retatrutide and Lipitor at once even though neither of us thought retatrutide was the main cause. Levels later fell to twice the upper limit. A Fibroscan showed no fatty liver. I may restart retatrutide later.

Good to hear the numbers are improving.
