I've been using 50mg of NAD+ every day for the past three to three and a half months. It took a couple months before I noticed a mild lift in energy and mood. It stayed pretty subtle the whole time. Out of curiosity I jumped to 250mg a day for the last three days and the difference is obvious. Energy feels smooth and steady through the whole day plus mood is clearly better. Has anyone else tried big jumps like that and felt the same thing?

I've thought about trying more. I know absorption isn't identical but I used to take 1000mg of a precursor daily for a long time before switching to the injectable form. Currently I'm at 150mg a day. Are you doing IM or SubQ? Curious what the stronger effect felt like at that higher amount.

250mg is a lot. I figured my occasional 100mg after two months was already on the high side. Since I like what it does for me I'm tempted to try more. A couple questions though: how do you handle dilution? I have 500mg vials. To get 250mg into a 1ml syringe I'd need to mix with 2ml of BAC. That seems like it would sting a lot or be hard to draw. Or do you split it into multiple shots?

I used to do IM but switched to injecting in the glute and it works fine.

I mix a 500mg vial with 5ml BAC so the 250mg dose comes out to 2.5ml in one shot. I put it in the glute and don't get any burning.

Are you putting the 2.5ml subQ into the fat above the glute or actually into the muscle? That's a wild way to take it.

It's subQ in the glute with a half-inch needle. Sometimes I only insert it halfway.

I've heard of people going up to 500mg subQ in one day for athletic events. It probably isn't that different from an IV drip of 750-1000mg over a couple hours except for how fast it hits.

I'm not very flexible so the only way I can reach my own glute is with a pen. Right now I'm using that for another peptide and it doesn't sting. I'll have to practice with regular syringes behind my back.

If it stings, adjust the pH with sodium bicarbonate using test strips to stop the burn.

Anything involving the glute makes me worry about hitting the sciatic nerve or an artery. For other injectables I prefer deep subQ or IM in the thigh muscle following standard video instructions.

In my first cycle with SS-31 and MOTS-C I used 150mg NAD+ on Monday Wednesday Friday. This summer I'm doing 100mg every day.

With higher or more frequent doses I would add daily oral TMG. I got a kit of NAD+ but I'm unsure how to use it or if it's buffered. I plan to reconstitute with 5ml but dosing and timing info online is all over the place.

Funny this came up because I just decided to try the same thing. Mixed 500mg with 2ml BAC and injected about 220mg. It definitely gave a strong kick.

I also increased today without any special reason, though not quite as high. I've been on 100mg three times a week for two weeks and went to 150mg today. Felt some flushing and had to sit down for a bit but the rest of the day was fine. I'll probably stick with 100mg and just do it more often. I reconstitute 1000mg with 2ml so my 150mg dose is only 30 units and I haven't had much stinging.

I push the plunger very slowly. It shouldn't burn unless you're in muscle instead of fat or the pH is off. I did it in the abdomen. Check pH if you have strips and keep it around 6 or lower. Let the dose warm to room temp in the syringe and pinch the skin hard so the poke is less noticeable. With MOTS-C I get some itchiness at the site for a while. The energy boost is great but I wouldn't go overboard because the strong feeling might be too much for some people.

Either you're using non-buffered NAD+ or I've just gotten used to it. I mix 500mg with 1ml BAC, draw 0.2ml subQ, and feel no sting at all.

No reliable blood test exists because NAD+ is unstable in blood and levels there don't reflect what's happening in organs and tissues. The same issue comes up with glutathione. The science supports that NAD+ is active, but whether cells can actually use it depends on mitochondrial health. I get a flush at 100mg but I'm not counting on big energy gains. Still, I'd like to try 500mg once. Typical IV doses are much higher. Both of us sometimes get stomach upset on injection days. I'm currently using 100mg three times a week. Clinics often start with a loading phase of 100mg then move to larger IV sessions and follow up monthly or weekly.

I'm taking 50mg NAD+. I don't think it does much by itself but I'm using it along with SS-31 and MOTS-C.

The oral precursors never gave me any noticeable effect. SubQ NAD+ does give a clear lift in energy and mood that lasts all day.

I tried 50mg three times a week with daily oral TMG for several months and felt nothing. Then I tried two weeks of 100mg daily and still nothing so I stopped. At 71 I wonder if I just don't have enough healthy mitochondria left for it to work. Same worry with BPC-157. Curious what people think about SS-31 and whether it would help with the other peptides.

SS-31 helped me. You need 4mg a day. Some say less is enough but I only got results at 4mg and I'm 54.

It was around 250mg. It happened because of a mixing mistake and pushing too fast. Every muscle seized for a few minutes and it was frightening. I thought about calling for help but figured it would be too late anyway.

If you need another IM site, look up ventrogluteal injection locations. It's a large muscle, safer for the concerns you mentioned, and reachable on your own.

I started TMG along with a couple other supplements as a way to support metabolism while in a calorie deficit. My energy has been surprisingly high and I've been sleeping like a log. Not sure if it's the stack or just the diet and training but some things are definitely working.

If you have a 500mg vial mixed with 5ml BAC and want 250mg, you'd need to split it into five separate 0.5ml syringes. That's a lot of injections.
