Klotho has one of the best names in biology — it's named after Klotho, the Greek Fate who spins the thread of life — and it earns it. It's a genuine "antiaging protein," with some of the most dramatic longevity data in all of aging science. The catch is a familiar one on this site: that data is mostly in mice, and "klotho injection" as a human therapy is far ahead of the evidence.

What klotho is

Klotho is a protein made mainly in the kidney and brain, first described in 19973. It comes in two forms: a membrane-bound version anchored to cells, and a soluble version — cut loose by enzymes and released into the bloodstream, where it acts like a circulating hormone2. That soluble, hormone-like form is the part most of the "antiaging" interest focuses on.

The mouse evidence is genuinely striking

This is what put klotho on the map. Mice bred to make too little klotho age rapidly and die young — the protein was literally discovered because a klotho-deficient mouse looked like a model of accelerated aging1. Flip it around, and mice engineered to make extra klotho live longer. That's *causal* evidence in an animal: change klotho, change lifespan. Findings that clean are rare in aging research, and they're the reason klotho is taken seriously.

What it actually does

Klotho isn't a mystery molecule. Its best-defined job is acting as a co-receptor for FGF23, a hormone that regulates phosphate and vitamin D — which is why the kidney is central to klotho biology2. The through-line: more klotho tends to go with healthier aging in humans5. But every one of these is an *association* — klotho levels correlate with better outcomes. That is a strong hint, not proof that raising klotho *causes* those benefits in people.

The honest gap: association is not therapy

Here's where the marketing gets ahead of the science. The mouse data is causal, the human data is associational — and the step everyone actually wants, giving klotho to people and measuring what happens, is still preclinical1.
